Abstract

Muguruma et al. developed a protocol for generating cerebellar organoids from human ESCs using FGF2, FGF19, and SDF1 to recapitulate isthmus organizer signaling and cerebellar plate formation. The organoids contained both Purkinje cell precursors and granule cell progenitors with layered organization, providing the first model for studying cerebellar development and ataxia disorders.
