Abstract
Lin et al. used CRISPR gene editing to generate isogenic APOE4/4 and APOE3/3 cerebral organoids, demonstrating that APOE4 drives increased amyloid-β accumulation, tau hyperphosphorylation, and cholesterol metabolism dysregulation independently of other genetic background effects. This Neuron 2018 study established the isogenic approach as the gold standard for studying APOE4 as an Alzheimer disease risk factor.
