Abstract

Miura et al. developed cortico-striatal assembloids by fusing cortical and striatal organoids containing DARPP-32+ medium spiny neurons, demonstrating functional cortico-striatal projections validated by optogenetic stimulation and calcium imaging. Published in Nature Biotechnology 2020, this Paşca lab protocol enables modeling of basal ganglia circuit disorders including Huntington disease.
