Abstract

Kim et al. demonstrated that ARVC-causing PKP2 mutations require metabolic maturation with lipogenic induction to manifest the disease phenotype in iPSC-CMs. Adult-like metabolic conditions trigger desmosomal dysfunction, abnormal lipid accumulation, and suppressed Wnt signaling, recapitulating key ARVC pathological features invisible in standard immature cultures.
