Abstract

Deuse et al. engineered hypoimmunogenic iPSC-CMs via triple CRISPR editing (B2M-KO, CIITA-KO, CD47-KI) that evade T-cell, NK cell, and macrophage killing. These universal donor cells survive allogeneic transplantation without immunosuppression in humanized mice, enabling off-the-shelf cardiac cell therapy products.
