Abstract

This protocol generates human gastric organoids (hGOs) through directed differentiation of human embryonic stem cells (hESCs) or human induced pluripotent stem cells (hiPSCs) into definitive endoderm, followed by stepwise induction of anterior foregut and antral stomach specification. The resulting three-dimensional organoids recapitulate gastric epithelial development in vitro and contain multiple gastric cell types including enteroendocrine cells, producing a physiologically relevant model of the human stomach antrum.

Definitive Endoderm Induction Days 0-3
  1. 1 Seed hESCs or hiPSCs for differentiation
  2. 2 Treat with Activin A to induce definitive endoderm
Foregut and Antral Specification Days 3-9
  1. 1 Apply Wnt, FGF4, and noggin to promote foregut fate
  2. 2 Transfer to 3D extracellular matrix with retinoic acid
  3. 3 Culture in EGF-rich medium to generate mature gastric organoids
Gastric Organoid Maturation and Maintenance Days 9-34+
  1. 1 Maintain organoids in 3D Matrigel culture
  2. 2 Monitor organoid formation and cellular differentiation
