Human Gastric Organoid Differentiation from Embryonic and Induced Pluripotent Stem Cells

Abstract
This protocol generates human gastric organoids (hGOs) through directed differentiation of human embryonic stem cells (hESCs) or human induced pluripotent stem cells (hiPSCs) into definitive endoderm, followed by stepwise induction of anterior foregut and antral stomach specification. The resulting three-dimensional organoids recapitulate gastric epithelial development in vitro and contain multiple gastric cell types including enteroendocrine cells, producing a physiologically relevant model of the human stomach antrum.
