Abstract

Demonstrated sequential introduction of the four major CRC driver mutations (APC, P53, KRAS, SMAD4) in normal human intestinal organoids using CRISPR/Cas9. Quadruple mutant organoids acquired niche-independent growth and tumorigenicity, recapitulating the classic adenoma-to-carcinoma progression sequence.
