Abstract
Established the first protocol for generating human colonic organoids (HCOs) from iPSCs by adding transient BMP4 signaling during hindgut differentiation. HCOs express colonic markers (SATB2, HOXD13) and lack Paneth cells, enabling region-specific modeling of colonic diseases like ulcerative colitis.
