Abstract

Visualized LGR5+ cancer stem cells in human colorectal cancer organoids using CRISPR knock-in reporters. Lineage tracing showed LGR5+ cells drive tumor growth, but their ablation revealed remarkable plasticity where LGR5-negative cells regenerate the stem cell pool, explaining resistance to targeted therapy.
