Tissue Acquisition and Processing Day 0 (within 4 h of collection)
  1. 1 Receive and document surgical or biopsy specimen
  2. 2 Mechanical dissection
  3. 3 Collagenase II digestion
  4. 4 Sequential straining, washing, and red cell lysis
  5. 5 Count viability and assess sample quality
Matrigel Dome Plating Day 0
  1. 6 Prepare Matrigel domes with PDAC cells
  2. 7 Plate domes and polymerise
  3. 8 Overlay with POM + ROCK inhibitor
Culture Monitoring and Establishment Day 1–14
  1. 9 Monitor daily and refresh medium
Passage, Expansion, and Cryopreservation Day 7–14 (first passage); then every 7–14 days
  1. 10 Recover organoids from Matrigel
  2. 11 Fragment organoids for passage
  3. 12 Re-embed in Matrigel and cryopreserve
