Phase 1 · Tissue Acquisition and Processing Day 0 (within 4 h of collection)
  1. a Receive and document surgical or biopsy specimen
  2. b Mechanical dissection
  3. c Collagenase II digestion
  4. d Sequential straining, washing, and red cell lysis
  5. e Count viability and assess sample quality
Phase 2 · Matrigel Dome Plating Day 0
  1. a Prepare Matrigel domes with PDAC cells
  2. b Plate domes and polymerise
  3. c Overlay with POM + ROCK inhibitor
Phase 3 · Culture Monitoring and Establishment Day 1-14
  1. a Monitor daily and refresh medium
Phase 4 · Passage, Expansion, and Cryopreservation Day 7-14 (first passage); then every 7-14 days
  1. a Recover organoids from Matrigel
  2. b Fragment organoids for passage
  3. c Re-embed in Matrigel and cryopreserve
