Abstract
This protocol describes the establishment and long-term culture of organoids derived from healthy oral mucosa and head and neck squamous cell carcinoma (HNSCC) patient tissue. The organoids recapitulate genetic, molecular, and morphological characteristics of the original tumors and retain tumorigenic potential in vivo, enabling in vitro drug screening for personalized treatment approaches.
